Comparing the production potential of IVD standard proteins in CHO cells and E. coli

Abstract

Raw materials for in vitro diagnostic (IVD) industry can be obtained from their natural sources but the recent development in recombinant protein technology has made them industrial-scale production more affordable. One important step in optimizing the manufacturing of recombinant proteins is the selection of suitable production platform. Available host systems include bacteria, mammalian cells, yeasts, insect cells and cell-free systems. The aim of this thesis was to compare the potential of two widely utilized host systems, Chinese hamster ovary (CHO) cells and E. coli, in the production of five recombinant proteins (NT-proBNP, cystatin c, cTnT, cTnI and NSE) that are used as reference materials in IVD immunoassays. The production potential was evaluated by experimenting the transient and stable expression of the selected proteins in CHO cells and comparing the results to data of similar products produced in E. coli. With expression tests in CHO cells, the production efficiency was found to be highly protein specific, suggesting that the molecular properties of proteins may guide the selection of a suitable host system. The achieved expression levels were utilized as a framework to create analysis tool to estimate the production cost of the selected standard proteins. By analyzing the expenditure profiles, E. coli fermentation proved its suitability in routine manufacturing. However, if the demand of a protein from the market is high, production in stable CHO cells demonstrated good adaptability due to the possibility to expand the production with already generated high-producing cells. Together with the capability of CHO cells to perform PTMs to express correctly folded and complex proteins with high functional properties, this finding supports their attractivity to be utilized as a production platform in IVD industry.

In vitro diagnostiikka -teollisuuden raaka-aineita voidaan eristää niiden luonnollisista lähteistä mutta rekombinanttiteknologian kehitys on tehnyt siitä vaihtoehtoisen tuottotavan käytettäväksi teollisuuden mittakaavan proteiinien tuotantoprosesseissa. Rekombinanttiproteiinien tuotannossa tuottosyteemin valinta on yksi tärkeä osa prosessin optimointia. Erilaisia tuottosysteemejä ovat esimerkiksi bakteerit, eläinsolut, hiivat, hyönteissolut sekä soluvapaatsysteemit. Tämän diplomityön tavoitteena oli verrata kahta yleisesti käytettyä tuottosysteemiä CHO soluja ja E. coli -bakteereita viiden rekombinanttiproteiinin tuotannossa, joita valmistetaan standardiproteiineiksi IVD vasta-ainetesteihin. Systeemien tuottopotenttiaalia verrattiin tutkimalla proteiiniekspressiota CHO soluissa transientilla ja stabiililla tuottomenetelmällä ja vertaamalla tuloksia samojen proteiinien tuottotasoihin saatavilla olevaan dataan E. coli -fermentoinnista. CHO soluilla tehdyillä ekspressiokokeilla havaittiin tuottotehokkuuden olevan proteiiniriippuvaista. Proteiinien erilaiset ominaisuudet saattavatkin ohjata sopivan tuottosysteemin valintaa erityisesti, jos tavoitteena on tuottaa sekretoituvia proteiineja. Saavutettuja tuloksia ekspressiokokeista käytettiin kustannusanalyysityökalun luomiseen, jonka avulla voitiin arvioida proteiinikohtaisia tuottokustannuksia. Työkalun avulla saadut kustannusprofiilit osoittivat E. coli fermentoinnin sopivuuden yleisesti tuottotavaksi tutkituille proteiineille. Havaittiin kuitenkin, että tuotteiden kysynnän kasvaessa stabiilituotto CHO soluilla on mukautuvainen kasvaviin tuottovolyymeihin, sillä jo valmistettuja tuottavia soluja voidaan siirtää lisää tuotantoprosessiin. CHO solut ovat myös kykeneviä valmistamaan post-translationaalisesti muokattuja proteiineja, jotka vaikuttavat niiden oikeaan laskostumiseen ja funktionaalisiin ominaisuuksiin. Nämä tekevät CHO soluista houkuttelevan vaihtoehdon käytettäväksi IVD teollisuuden prosessien tuottosysteeminä.
