Functional expression of the carbon monoxide methylating acetyl-CoA synthase enzyme complexes and their β subunits from alkanotrophic archaea in Methanosarcina acetivorans and in Escherichia coli.

Abstract

The metabolic systems, with which some alkanotrophic archaea can metabolise longer alkanes like ethane, propane, and butane, remain a subject of study. Divergent CODH/ACS enzyme complexes are hypothesised to enable the metabolising of these substances. Due to their reversible nature, these complexes have sparked interest as tools for the synthesis of longer alkanes. This thesis aims at the functional expression of CODH/ACS complexes and their ACS subunits found in different archaea, for use in in vitro and in vivo assays. For in vitro assays, the ACS subunits of Ca. S. butanivorans and Ca. E. thermophilum were expressed and purified for the first time. In addition, the genes for the CODH/ACS complexes from M. acetivorans, Ca. S. caldarius, Ca. S. butanivorans and Ca. E. thermophilum were cloned into a new shuttle vector pGA2 and the vectors were transformed into an archaeal host to perform an in vivo growth assay. The first time ACS subunit purifications were successful for both Ca. S. butanivorans and Ca. E. thermophilum. The enzyme yields for Ca. S. butanivorans ACSs were significant, reaching up to 2.6 mg of enzyme per a litre of culture. Yields for the ACS of Ca. E. thermophilum were lower, reaching 0.42 mg of enzyme per litre of culture, despite attempts to improve the expression and purification process. The transformation of M. acetivorans CODH/ACS containing vector into the archaeal host MCD21 was performed successfully. The cloning and transforming of other CODH/ACS complexes remain subjects of future experiments.

Metaboliset systeemit, joilla jotkin alkanotrofiset arkeonit hajottavat pidempiä alkaaneja, kuten etaania, butaania sekä propaania, ovat jatkuva tutkimuksen aihe. Poikkeavien CODH/ACS entsyymi kompleksien on hypotesoitu mahdollistavan näiden aineiden hajottamista. Nämä kompleksit ovat herättäneet kiinnostusta alkaanien syntetisoinnin työkaluina, niiden reaktioiden käänteisyyden takia. Tämän diplomityön tavoite on eri arkeonien CODH/ACS kompleksien ja ACS-entsyymien funktionaalinen ekspressio in vitro ja in vivo analyyseja varten. In vitro analyysejä varten ACS-entsyymiä Ca. S. butanivoransista ja Ca. E. thermophilumista eristettiin ja puhdistettiin ensimmäistä kertaa. Lisäksi CODH/ACS geenit M. acetivoransista, Ca. S. caldariuksesta, Ca. S. butanivoransista ja Ca. E. thermophilumista kloonattiin uuteen pGA2 plasmidiin ja nämä plasmidit transformoitiin arkeoni-kasvualustaan in vivo kasvuanalyysejä varten. Ensimmäistä kertaa tehdyt Ca. S. butanivoransin ja Ca. E. thermophilumin ACS entsyymin pudistukset onnistuivat. Saannot Ca. S. butanivoransin ACS-entsyymien kohdalla olivat korkeita, saavuttaen parhaillaan 2.6 mg entsyymiä kasvatuskulttuurilitraa kohden. Saannot Ca. E. thermophilumin ACS entsyymin kohdalla olivat alhaisempia ja paras saavutettu saanto oli 0.42 mg entsyymiä kasvatuskulttuurilitraa kohden. M. acetivoransin CODH/ACS geenit sisältävä plasmidi transformoitiin MCD21 kasvualustaan onnistuneesti. Muiden CODH/ACS geenien kloonaaminen plasmideihin ja näiden plasmidien transformaatiot jäävät tulevaisuuden kokeiden kohteeksi.
