Abstract
Rahkan ja raejuuston sivutuotteena syntyy hapanta heraa, jota käytetään pääasiallisesti eläinten rehuna. Käänteisosmoosilla konsentroidun happaman heran korkea sokeripitoisuus parantaa sen käyttöpotentiaalia myös etanolituotannossa. Työn tavoitteena oli kerätä esitietoja jatkuvatoimisen etanolintuottoprosessin suunnitteluun. Tässä työssä tutkittiin voidaanko immobilisaatiofermentoinnilla ja nanosuodatuksella tehostaa etanolintuotantoprosessia. Tavoitteena oli maksimoida etanolin volumetrinen tuottavuus säilyttäen herassa kaupallisesti kannattavalle jälkikäsittelylle vaadittava etanolipitoisuus. Nanosuodatuksella pyrittiin konsentroimaan fermentoinnin jäännöslaktoosi sekä saavuttamaan mahdollisimman matala retentio etanolille ja konsentraatin osmoottista painetta nostaville komponenteille. Tarkoituksena oli tuottaa retentaattia, jota voisi kierrättää takaisin fermentointiin. Työhön valittiin laktoosia hyödyntävä Kluyveromyces marxianus VTT C-75025 -hiiva. Hiivan etanolituotantokyky ja raaka-aineen esikäsittelytarve varmistettiin ravistelupullofermentoinneilla ennen immobilisaatiofermentointia. Esikokeiden tulosten perusteella hapan heratiiviste päätettiin saostaa lämpökäsittelyllä ennen fermentointia. Immobilisaatiofermentoinnit suoritettiin sekoitussäiliö- ja pakattu peti -reaktoreissa haapahakkeeseen immobilisoidulla hiivalla. Fermentoinneilla saavutettiin 0,50 g/(kg h) volumetrinen tuottavuus ja 27,0 g/kg etanolipitoisuus 0,0192 h-1 laimennusnopeudella. Koetta häiritsivät kontaminaatiot syöttösäiliössä ja fermentoreissa. Raaka-aine vaatii prosessilta parempaa kontaminaatioriskin hallintaa. Työssä suoritettiin suuren mittakaavan panosfermentointi, jolla saavutettiin 53,9 g/kg etanolipitoisuus 78 % saannolla kuluneista sokereista ja maitohaposta. Panosfermentointi on immobilisaatiofermentointia kypsempi tekniikka heran etanolituotannossa. Nanosuodatuksessa selvitettiin konsentrointisuodatukseen soveltuvat operointiparametrit ja kalvotyypin merkitys. Synder Filtrationin NFX ja Koch Membrane Systemsin SR3D toimivat parhaiten. Konsentrointisuodatuksessa saavutettiin 2,10 tilavuusperusteinen konsentrointikerroin, jonka jälkeen suodatusliuos saostui. Valituilla kalvoilla laktoosiretentiot olivat suodatuksen ajan yli 99 %. Suodatuksessa saavutettiin negatiivinen etanoliretentio, joka tarkoittaa permeaattiin päätyvän suuremman etanolipitoisuuden kuin retentaattiin. Karboksyylihapot ja tietyt suolat konsentroituvat nanosuodatuksessa, joka aiheuttaa epävarmuutta retentaatin uusiokäytössä. Suolojen retentioiden määritys ja negatiivisen etanoliretention erotusmekanismin selvittäminen vaatii jatkotutkimuksia. Prosessi tulisi lisäksi simuloida, jotta voitaisiin määrittää realistiset prosessivirtojen koostumukset.

Acid whey is a by-product in cottage cheese and quark production. It is mainly utilized as animal feed. Acid whey concentrated by reverse osmosis has a large sugar content, which enables the use of this fraction in ethanol fermentation. The goal of this thesis was to collect preliminary information for a continuous ethanol production process. The purpose of this study was to intensify whey based ethanol production by yeast immobilization and nanofiltration. The goal of this work was to maximize the volumetric productivity of ethanol, while keeping the ethanol concentration on an acceptable level. Nanofiltration was used in order to concentrate the residual lactose in the fermentation, as well as obtain a low retention to ethanol and other components that increase the osmotic pressure of the retentate. The goal was to produce a retentate that could be recycled back to the fermentation. Yeast Kluyveromyces marxianus VTT C-75025 was chosen for the experiments. Ethanol production and pretreatment requirements were tested in shake flask cultivations. The results indicated that heat treatment was necessary in order to precipitate calcium phosphate out from the concentrated acid whey. Fermentations with immobilized cells were carried out in a continuous stirred-tank re-actor and a packed bed reactor. The yeast was immobilized on aspen chips. A volumetric productivity of 0,50 g/(kg h) and a final ethanol titer of 27,0 g/kg was achieved with the dilution rate of 0,0192 h-1. Contaminants in the feed tank and reactors disturbed the experiment. Based on the observations made during the experiments, it can be concluded that the raw material requires better methods to reduce contamination risks. In comparison, a large scale batch fermentation was carried out. It gave an ethanol titer of 53,9 g/kg with 78 % yield from sugars and lactic acid. Batch fermentation is more mature as a technique than immobilized cell fermentation in ethanol production from whey. Operation parameters and the impact of the membrane type were examined in the preliminary nanofiltration tests for concentration. NFX from Synder Filtration and SR3D from Koch Membrane Systems gave the best performance. A volume reduction factor of 2,10 was achieved before the retentate precipitation. The retention of lactose was over 99 % during the concentration. Negative ethanol retention was obtained in the experiment, which means that the permeate had a higher ethanol concentration than the retentate. Carboxyl acids and certain salts are concentrating during nanofiltration, which will cause uncertainty to the re-use of retentate. Further research is required in order to determine the salt retentions and the separation mechanism behind the negative ethanol retention. Furthermore, the process should be simulated in order to determine feasible process stream compositions.
