Handheld FTIR for quantification of degree of degradation of drugs during pharmaceutical cleaning verification

Abstract

Ensuring the cleanliness of equipment and facilities is an important process in the pharmaceutical manufacturing industry to prevent cross-contamination between products and maintain product quality. Conventional cleaning verification involves collecting samples which are subsequently sent to off-site laboratories for analysis. However, these conventional methods often prove time-consuming and may delay production. This has led to the exploration of analysis methods that can be performed with a portable device in the production facility and can analyze the equipment surface directly, known as real-time cleaning verification.

This thesis describes a method for real-time quantification of the degree of degradation of drugs for cleaning verification using a handheld FTIR device. In order to utilize the FTIR device for this purpose, chemometric models which could predict the degree of degradation of samples based on their FTIR spectra were developed. The chemometric prediction models were constructed by utilizing PLS R (Partial least squares regression) and PLS-DA (Partial least squares discriminant analysis) based on reference values of the degradation level and spectra of the samples. Additionally, this thesis evaluated whether the results obtained using the handheld device are subject to sampling bias. Specifically, the contribution of collection mode to sampling bias was investigated. This was achieved by statistically comparing the results of FTIR analyses conducted in different settings.

In this thesis a successful PLS-R model was developed. This model was able to predict the degree of degradation of API samples. Additionally, a PLS-DA model which was able to predict a classification of degraded or non-degraded based on the spectrum of a sample is presented. The evaluation of the sampling bias indicates that analyses may be subject to bias based on the analysis situation.

This thesis provides new insight in producing prediction models for use in real-time cleaning verification as it includes the degree of degradation of an API in an environment containing cleaning agent residues.

Att säkerställa renheten av utrustning är en viktig process inom läkemedelstillverkningsindustrin för att förhindra korskontaminering mellan produkter och för att upprätthålla produktkvaliteten. Konventionell verifiering av rengöring innefattar insamling av prover, som sedan skickas till laboratorier utanför produktionsanläggningen för analys. Dessa konventionella metoder är dock ofta tidskrävande och kan förorsaka dröjsmål i produktionen. Detta har lett till ett behov av analysmetoder som kan utföras med en portabel enhet i produktionsanläggningen och som kan analysera utrustningens yta direkt.

Denna avhandling beskriver en metod för kvantifiering av nedbrytningsgraden av läkemedel för renhetsverifiering i realtid med hjälp av en portabel FTIR-enhet. För att kunna använda FTIR-enheten för detta ändamål utvecklades kemometriska modeller som kunde förutspå nedbrytningsgraden av läkemedlet utgående från dess FTIR-spektrum. De kemometriska modellerna utveckades genom (Partial least squares regression) och PLS-DA (Partial least squares discriminant analysis)- analys baserat på provernas spektra samt ett referensvärde av nedbrytningsgraden. Utöver det utvärderade denna avhandling ifall resutaten av analyser är föremål för ett systematiskt fel på grund av omständigheterna kring analyssituationen, såkallad provtagningsbias. Specifikt undersöktes analyssituationens kontribution till provtagningsbiasen. Detta åstadkoms genom statistisk jämförelse av resultaten från FTIR-analyser som utförts i olika analyssituationer.

Avhandlingen presenterar en PLS-R-modell som kan förutsäga nedbrytningsgraden av API-prover. Dessutom presenteras en PLS-DA-modell som kan förutsäga en klassificering av nedbrutna eller icke-nedbrutna prover baserat på provets spektrum. Utvärderingen av det systematiska felet som uppkommer av analyssituationen indikerar att analyser kan vara föremål för ett systematiskt fel baserat på analyssituationen.

Denna avhandling ger ny insikt i att skapa modeller för användning i rengöringsverifiering i realtid, eftersom den inkluderar graden av nedbrytning av en API i en miljö som innehåller rester av rengöringsmedel.
